When I first started this game I didn't realize just how important runes are. Runes can actually make or break your monsters. That being said you want to be pretty careful about what you do with them. Why? Because if you goof it generally means you have to spend money to remove the runes you don't want. There are the free rune removal days, but they happen once a month (I think) and you never really know when that will be.
The GENERAL; and I do mean that, rule of thumb when low to mid level is this:
If it is listed as an ATTACKER type monster: Use Fatal and Blade Runes opting for ATK% bonuses in slots 2/4/6.
If it is listed as SUPPORT type monster: Use Swift and Energy Runes opting for SPD and then HP% bonuses.
If it is listed as a DEFENSE type monster: Use Guard Runes and go for DEF% runes in slots 2/4/6.
If it is listed as an HP monster: Use Energy Runes and go for all HP% bonuses.
There are many exceptions to these rules and it really depends upon the monsters. Sometimes certain monsters abilities are based on a certain stat. An example of this is the Water Fairy Elucia. Even though she is a SUPPORT monster, if you read the description of her second skill which is her healing one you will see it is directly related to her Attack Power. So for her you would want Fatal Runes with ATK% and then perhaps either Energy runes for HP% early on in the game.
If all else fails and you aren't sure you can ask us. We have your best interest at heart. Asking random people in a chat room in the game could lead you down the wrong road. Some people just like messing with other people. Take advice there with a grain of salt.
